In vitro rooting and acclimatization of Corymbia citriodora seedlings

The objective of this study was to evaluate the effect of indolbutyric acid (IBA) concentrations on in vitro rooting of Corymbia citriodora juvenile shoots, and sealing forms on the acclimatization of micropropagated seedlings. For rooting, shoots were placed in a culture medium with four concentrations of IBA (0, 1.0, 2.0, and 4.0 mg L-1). In pre-acclimatization, seedlings germinated in vitro and without contamination were transplanted into polyethylene cups containing substrate. The cups were packed in plastic bags for 10 days and kept in a culture room. Subsequently, three treatments were applied: no coverage; intact plastic bag, and perforated plastic bag. In acclimatization, the history of pre-acclimatization treatments was maintained. The seedlings were transplanted into tubes containing substrate and kept initially in a greenhouse and then in a shade house. The results demonstrate that concentrations of 1.0; 2.0 and 4.0 mg L-1 of IBA were efficient in the rooting of juvenile explants of C. citriodora, and the highest concentration used provided the highest average number of roots. The absence of cover in pre-acclimatization is recommended for greater survival rate, height, and dry mass of seedlings in the acclimatization stage.
